For installation and training, we would need the following requirements: - Space: 40 cm wide x 51 cm deep x 75 cm high. The system can be set up outside a fume hood. The exhaust gases from the collector need to be diverted to the air venting system. - Power: 1 x 220 V outlet - Solvent: Doesn't have to be HPLC-grade, but mostly particle-free: cyclohexane or similar, ethyl acetate, and any other solvents for your runs/samples. - In case we want to work in reversed phase: water and ACN/MeOH. I have a C18 test column with me that we can try out. - Consumables: Tissue paper, 2 ml and/or 5 ml syringes, needles - Waste: A waste container (e.g., a 2.5 L bottle or canister) - Test tubes: 9 mm, preferably a whole carton - For the iELSD, we need a small amount of HPLC-grade isopropanol or ethanol (250 ml or 500 ml is sufficient for now) - iELSD gas: The gas tubing has a 6 mm OD. We would need a corresponding adapter at the gas outlet, which should also have a pressure gauge. The line requirements are very low: 1.2 bar and 1 L/min are sufficient. For the training: - We want to make the training as realistic as possible. It would therefore be ideal if you could prepare some real samples. Attached you will find the loading table, which I will explain again during the training. Between 50 mg and 1 g of sample can be loaded onto an F0012 cartridge, with the load depending largely on the difficulty of the separation problem (see DeltaCV). The software helps with the selection. For training, sample quantities of 100 mg to 400 mg would be perfect. - A suitable TLC for the respective sample is also welcome, with values ideally between Rf 0.1 and Rf 0.35 (or at least one of these values). - You are welcome to download the free app "TLC to Flash and Prep" in advance. With the app, we can simply take a photo of the TLC and send it to the device via Bluetooth. For normal-phase separations, many samples are not well soluble under the initial conditions of the run (high proportion of the less polar solvent). In this case, a dry load is recommended. I will bring a few test cartridges so that we can run through the process during the training. For the DryLoad, we need: - Sample bonded onto silica/Celite/Isolute in a ratio of 1 part sample to 2 parts Celite/silica - Some material to fill the DryLoad cartridge. An old wash bottle filled with sea sand is ideal. If you cut off the top of the syringe, you can simply pump the sea sand into the DryLoad to fill it up. - A small tripod with a clamp for filling the DryLoad is helpful We look forward to the training and installation.
